What is KPV?
KPV is a naturally occurring tripeptide consisting of lysine, proline, and valine. It represents the C-terminal active fragment of alpha-melanocyte-stimulating hormone (alpha-MSH), the portion of the larger hormone responsible for its anti-inflammatory activity. What makes KPV particularly interesting to researchers is that it retains the anti-inflammatory properties of alpha-MSH without the pigmentation effects associated with the full-length hormone. This selectivity has made KPV one of the most studied small peptides in inflammation and gut health research.
Mechanism of Action
KPV exerts its effects primarily by entering cells and interfering with inflammatory signalling pathways at the intracellular level. Research indicates it inhibits the activation of NF-kB, a key transcription factor that drives the production of pro-inflammatory cytokines. By suppressing this pathway, KPV reduces the downstream release of inflammatory mediators including TNF-alpha and IL-6. Notably, KPV is transported into cells via the PepT1 transporter, which is highly expressed in inflamed intestinal tissue, meaning the peptide preferentially accumulates where inflammation is greatest.

KPV has been studied across a range of inflammatory and healing contexts. Key areas include inflammatory bowel disease and colitis models, where its PepT1-mediated uptake in inflamed gut tissue is particularly relevant. It is also researched for skin inflammation and wound healing, where its anti-inflammatory activity supports tissue repair. Additional research explores its antimicrobial properties, its role in reducing systemic inflammation, and its use in combination with other repair peptides such as BPC-157 and TB-500 in blended formulations.
